چکیده

Although leukocytes from all 13 acute lymphoblastic ieu kemia patients examined had high terminal deoxynucleoti dyl transferase (terminal transferase) activity (20 to 100 units/mg of cellular DNA, where 1 unit equals 1 nmole of nucleotide polymenized in 1 hr) and those from 21 acute myelocytic leukemia patients had low terminal transferase activity (0.2 to 2 units/mg of cellular DNA), the bone marrow and peripheral blood leukocytes from 2 patients with acute myeIocytic leukemia, diagnosed on the basis of clinical features and the morphology, cytochemistry, and cytoge netics of the leukemic cells, had terminal transferase activ ity (39 to 52 units/mg of cellular DNA) equivalent to that found in leukemic lymphoblasts. These results bring under question the specificity of high terminal transferase activity outside of the thymus as a marker for leukemic lympho blasts and, secondarily, the derivation of acute lymphoblas tic leukemia cells in all cases from thymocytes. Perhaps malignant transformation in a pleunipotent stem cell with derepression of the genome for terminal transferase could account for high terminal transferase activity observed in certain leukemic cells.

Terminal deoxynucleotidyl transferase as a biological marker for human leukemia.

High levels of terminal deoxynucleotidyl transferase have been observed in leukocytes of 7 out of 20 patients with chronic myelogenous leukemia in acute blast phase of the disease. These levels are comparable to the levels observed in human and calf thymus gland and cell lines with some T cell characteristics (Molt 4 and 8402). Negligible levels of this activity were observed in chronic myeloge...

Terminal Deoxynucleotidyl Transferase Activities in Human Blood Leukocytes and Lymphoblast Cell Lines: High Levels in Lymphoblast Cell Lines and in Blast Cells of Some Patients With Chronic Myelogenous Leukemia in Acute Phase

Terminal deoxynucleotidyl transferase, an enzyme which catalyzes the polymerization of deoxyribonucleoside triphosphates, elongating oligoor polydeoxynucleotide chains, but without direction from a nucleic acid template, is thought to be specific for thymus gland and thymusderived cells. We have confirmed the observations that high levels are characteristic of thymus gland with both human and c...

Track detection on the cells exposed to high LET heavy-ions by CR-39 plastic and terminal deoxynucleotidyl transferase (TdT)

Background: The fatal effect of ionizing radiation on cells depends on Linear Energy Transfer (LET) level. The distribution of ionizing radiation is sparse and homogeneous for low LET radiations such as X or γ, but it is dense and concentrated for high LET radiation such as heavy-ions radiation. Material and Methods: Chinese hamster ovary cells (CHO-K1) were exposed to 4 Gy Fe-ion 2000 keV/...
